The Science of Hyaluronic Acid for Skin: Beyond the Hydration Hype
Hyaluronic acid skincare benefits have evolved far beyond simple hydration.
Hyaluronic acid is an important component of the extracellular matrix, with loss starting at 25 years old.
This naturally occurring glycosaminoglycan has become the cornerstone of effective anti-aging formulations, offering clinically proven results that go deeper than surface-level moisture.
For 18 years, Marianella has been handcrafting formulations in Brooklyn that harness hyaluronic acid's true potential. Drawing from three generations of Venezuelan botanical beauty wisdom, our approach to this celebrated ingredient reflects both scientific rigor and artisanal expertise.
What Is Hyaluronic Acid in Skincare?
Hyaluronic acid is a naturally occurring glycosaminoglycan found throughout the body's connective tissue. Glycosaminoglycans are simply long unbranched carbohydrates, or sugars, called polysaccharides. HA is the main component of what gives your skin structure, and is responsible for that plump and hydrated look.
Unlike many skincare ingredients that work on the surface, hyaluronic acid operates at the cellular level.
In humans, almost half of the total HA concentrates in the skin, but its availability decreases with age.
This natural decline explains why incorporating quality hyaluronic acid products becomes increasingly important for maintaining skin health.
The Molecular Weight Factor
Not all hyaluronic acid delivers the same results.
The size of HA appears to be of critical importance because the permeability of HA is predominantly related to its molecular weight. HMW‐HA has very limited permeability through the skin and primarily stays on the skin's surface, forming a thin protective hydration layer, while LMW‐HA can permeate the stratum corneum, epidermis, and deeper dermal layers.
HA with a low molecular weight of 20–300 kDa passes through the stratum corneum, while high molecular weight (1000–1400 kDa) HA is largely impermeable.
This explains why sophisticated formulations combine multiple molecular weights for comprehensive benefits.
Clinical Evidence for Hyaluronic Acid Benefits
Recent studies demonstrate impressive results for hyaluronic acid for skin improvement.
A RCT including 40 females with mild‐to‐moderate clinical signs of skin aging demonstrated improvement in skin appearance versus placebo after 30 days of using six types of topical HA of different molecular weights according to wrinkle volume.
Topical application of all 0.1% HA formulations used in this study led to significant improvement in skin hydration and elasticity. Application of low-molecular-weight (LMW) HA was associated with significant reduction of wrinkle depth, which may be due to better penetration abilities of LMW HA.
The specific improvements are measurable.
After one month, they found that treatment with 130 kDa HA was the most effective, increasing skin elasticity by 20 percent. Both the 50 and 130 kDa groups had significant improvement in wrinkle-depth and skin roughness after 60 days.
Penetration and Efficacy Data
The efficiency of penetration was greater for the lower molecular weight molecules, with hydrolyzed glycosaminoglycans 400 Da, sodium hyaluronate 800 Da, HA 1 kDa, hydrolyzed sodium hyaluronate 5 kDa, sodium hyaluronate 20 kDa, hydrolyzed HA 50 kDa, sodium hyaluronate 80 kDa, and sodium hyaluronate 100 kDa showing penetration of 14% to 19% and larger molecules displaying penetration of 2.73% to 10.2%.

The Water Retention Reality
While marketing claims often state that hyaluronic acid holds 1000 times its weight in water,
I found that hyaluronic acid can only hold up to 50x its weight in water. Perhaps more egregious than the thoroughly false claim, is that there is not an established way in the literature to determine how much hyaluronic acid actually absorbs.
HMW HA saturated at ~6% (≈30× water:HA, w/w). LMW HA saturated at ~5% (≈20× water:HA, w/w).
The genuine benefits of hyaluronic acid extend far beyond simple water retention, encompassing cellular signaling, barrier function, and structural support.
Optimizing Hyaluronic Acid Application
HMW‐HA has a positive effect on hydration of upper epidermis layers, which translates into a lower transepidermal water loss.
For maximum efficacy, apply hyaluronic acid serums to damp skin, then seal with appropriate moisturizers.
The concentration of HA in cosmetics typically ranges from 0.2% to 1%.
Higher concentrations don't necessarily mean better results. Professional formulations balance concentration with molecular weight distribution and complementary ingredients.
The Future of Hyaluronic Acid Skincare
Cosmetic applications of LMW and hybrid HA included treatment of skin ageing, enlarged pores, and skin laxity, as well as enhancement of skin hydration. Topical LMW-HA administration promoted healing after cosmetic procedures, chemical peels, and ingrown toenail surgery.
Emerging research explores combination therapies and novel delivery systems.
HA in cosmetic products is often combined with other bioactive ingredients, particularly amino acids, peptides, vitamins, plant extracts, and probiotics, to enhance its effect or provide additional benefits for the skin.
